Specifically, how extended would it take to drive from Lower Manhattan (Greenwich Village area) to JFK airport on a weeknight around or after midnight? What kind of traffic should I want to encounter?
I would give yourself an hour but you will indubitably be there in 20-30 minutes. Traffic shouldn't be a problem.

I am customary to New York close to Christmas and will be staying at a hotel near or in midtown & was wondering which of these two airports would be more suitable to fly into. Thanks in advance!
LaGuardia is closer, but doesn't have superb mass transit connections. There is no rail stop there at all. Just buses, though there are subways a very blunt bus ride away.
JFK is farther, but has a highly-developed mass transit system built for the particular purpose of getting air travelers into the city. They call it the "AirTrain." From the AirTrain, you can relate to the A, E, J, and Z subways, as well as the Long Island Rail Road.
Consider Newark Liberty as well; this might indeed be MOST convenient, with well-developed mass transit connections. It's also VERY close to Manhattan.
So, closer isn't necessarily more within easy reach.
How Much Cost A Taxi From Bus Station 42st Manhattan To JFK Airport?

i have to take a soaring at JFK im comin from Pensilvannia so my bus gonna leave me at bus station 8av and 42st Manhattan and i need transportation to the airport anybody remember what i can do ?
$45 addition tip for the taxi. Whew!
If you're open to other options, you have a few:
A short hike (or subway ride) down 8th Avenue is Penn Spot at 33rd Street (one stop away on the 1,2,3,A,C,E subway lines). There, you can get the Long Cay Rail Road. Take it to Jamaica (the majority of LIRR trains pass through there), and get the AirTrain to JFK. Sufficiently quick trip, might even rival or exceed the speed of the taxi if the transfers are timed justified right.
Or, at the bus terminal, you can go down and get a Queens-bound E subway ($2); it'll run express through Queens and take you to Jamaica, put to the AirTrain ($5). The E comes frequently enough (every 5 or 10 minutes or so?), and the AirTrain comes every 2 to 8 minutes.
So, you have a few options, let me certain if you want more info on any of them. Consider time as a factor. If you have a good, comfortable amount of time, I absolutely recommend either the E train or the LIRR to the AirTrain. Take your amount of luggage into consideration as well. The mass transit options do have all the resulting ramps and elevators to make it comfortable, though. I've comfortably carried large television cameras on the LIRR and tube. That said, traveling with luggage to the airport is one of the VERY few situations in which I'd THINK about taking a taxi, though I'd still be very tempted to go legions transit if it was to JFK. LaGuardia's another story.

I'll be in NY for a week, I'll dismount in JFK airport and my hotel in in lower east side. I would like to use mainly the subway.
I know about bus service, but I would submit enter NYC from the undreground (subway) or by train.

What Is The Best Way To Go To Port Authority Bus Stop In Manhattan From JFK Airport In The Morning?

I am arriving JFK major-domo airport in the morning by a redeye flight. I will be travelling furhter from Port Authority toward NJ suburbs.
my counsel as a native new yorker is to take a shuttle or taxi, taxi could be pricey, the shuttle a little cheaper. of line public transportation would be the cheapest option but not that much cheaper then a shuttle especially if you have luggage i would sidestep public transportation in NYC. If you do choose to take the cheapest form which would be public transportation you have to take the Air Train from your ending to the Sutphin Blvd. (Jamaica Station) E train station. Then catch a manhattan bound (crowd trade center bound) E train and get off at 42nd street/port authority.
IN ADDITION: I neglected to blab about you that on saturday and Sunday the E train does not travel along the E route on the weekends due to construction, it globe-trotting trips down the F train route which means that you will not be able to reach port authority station via the E job on a saturday or sunday, instead you must that the Air train to a Manhattan bound A train, which will be longer because it doesnt run verbalize.
