I will be doing the subway to towers walk/run in from Brooklyn to Downtown Manhattan on September 28. I am trying to find out what train I will take from 34th street to Downtown Manhattan around Chambers roadway?
That depends where in 34th St you end.
A/C's Chambers St, World Trade Center on E, 2/3 on Park Place is connected.
Senate St/West Broadway is 2/3 train.
R and W train is at City Hall, which is few blocks north to Chambers St.
Brooklyn Link on 4/5/6 is connected to Chambers St station on J/M/Z.

I wish for to get both of my nipples pierced and I am looking for a reputable piercing parlor in downtown Manhattan. Preferably on the west side in the Greenwich Village close. Can anyone recommend one?
You miss to go to either NY Adorned on 2nd Ave (a few blocks north of the Whole Foods, around 3rd st?) or Venus Modern Body Arts. Keep off the creepy places on West 4th. St. Marks isn't where you want to be either. Honestly, I wouldn't trust any group that isn't Venus or NY Adorned.

Besides subways, anyone knows what bus most suitable to get to downtown Manhattan (Liberty Plaza) best for weekdays in the morning?
Where are you starting from?
Don't take the bus. During to-do hours street traffic will slow you down to a crawl. The subway would be faster and runs more frequntly.
But if you quite want to do it by bus - if you are in Manhattan, the M1 and M6 bus runs down Broadway and stops either in front of the building or a block away.
